Analysis of the visible spectra of some sperm-whale ferrimyoglobin derivatives.

چکیده
The visible spectra of sperm-whale metmyoglobin and its fluoride, hydroxide, cyanide, azide, cyanate, formate, acetate, thiocyanate and nitrite derivatives were measured and resolved into Gaussian components. A linear correlation between the intensity of the band at 18500cm.(-1) and the magnetic susceptibility is found, suggesting that the band is the beta-band of the low-spin form. The analysed intensity of this band can be used to determine the relative proportions of the two spin states present. The spectra of the pure high-spin and low-spin components of each derivative can be reconstructed. Correlations between the energy of the near-infrared band and the visible bands of the high-spin form, and between the position and intensity of the 16000cm.(-1) band, are demonstrated. The relationship between the position of the Soret band and the magnetic susceptibility can be explained in terms of the equilibrium between spin states.

Absorption spectra of sperm-whale ferrimyoglobin.
1. Sperm-whale ferrimyoglobin was found to contain 0.308% of Fe, on a dry weight basis, corresponding to a molecular weight of 18130. The solid takes up moisture to an equilibrium state, and was then assayed to contain 0.280% of Fe. 2. Absorption spectra are presented for acidic ferrimyoglobin, Fe(+)(H(2)O), and its conjugate base, Fe-OH, as well as for the fluoride and cyanide complexes, withi...
متن کاملThe Eguilibrium Unfolding Parameters of Horse and Sperm Whale Myoglobin
The urea and guanidine hydrochloride (GuHCI) unfolding of sperm whale ferrimyoglobin and of horse ferrimyoglobin has been studied under equilibrium conditions at both neutral and acid pH. Using two spectral parameters (the Soret absorbance and the molar ellipticity at 222 nm) to monitor conformational equilibria, and various thermodynamic treatments of the resulting data, it was found that the ...
متن کاملSome interrelations between carboxymethylation and heme reactions in sperm whale myoglobin.
The effect of carboxymethylation of sperm whale ferrimyoglobin on the conversion of the protein to the deoxymyoglobin (ferromyoglobin), oxymyoglobin, and carboxymyoglobin forms was studied. Judged by observations on the absorption spectra and on optical rotation at 233 rnp, the conversions to the other heme states were largely unaffected by carboxy methylation to the extent of an average of 7 o...
متن کاملIsolation of sperm whale myoglobin by low temperature fractionation with ethanol and metallic ions.
1. A method is described for the isolation and purification of myoglobin from sperm whale skeletal muscle. The aqueous extract is brought to 25 % ethanol by volume at 10 to lZ” and pH 6.5. A fraction precipitated with lead subacetate is discarded. Myoglobin is precipitated from the supernatant solution at pH 7.20 by adding zinc acetate to a concentration of approximately 10 InM. The precipitate...
متن کاملComparison of myoglobins from harbor seal, porpoise. and sperm whale. VI. Renaturation of copper(II)-denatured ferrimyoglobins.
Denaturation of dilute solutions of ferrimyoglobins at pH 650 and 50’ by a 7to lo-fold molar excess for copper(D) is completely reversible on addition of excess chelating agent. The renaturation reaction exhibits first order kinetics. In the ultracentrifuge, the copper(D)-denatured proteins show little aggregation except for an increase in sedimentation constant for the sperm whale protein from...
